▶ 강의제목 : Noncanonical function of a new inflammatory cytokine IL-32: 2005-2016

 IL-32 has been reported as NK 4 transcript that produced in T cell or NK cell and thought to be secreted because of the presence of signal sequence and the lack of transmembrane region, but its function has remained unclear. Recently, IL-32 has been redefined as a proinflammatory cytokine, and it is induced in epithelial cells and monocytes by IFN-γ stimulation in a time-dependent manner (IL-32: a new cytokine and inducer of TNFα. Immunity 22(1), 131, 2005).
Nine isoforms of IL-32 by alternative splicing have been reported (Biochimie, 2014), but functional differences among isoforms have not yet been elucidated. There are many uncertain and doubtful points regarding on IL-32: IL-32 is hardly detected in culture media; IL-32 has no signal sequence; IL-32 has too many isoforms; IL-32 isoforms are localized and interacted each other intracellularly: there is no IL-32 gene in mice and IL-32 receptor has not been reported yet.

In order to investigate the non-cannonical function of isoforms, we focused on the following areas:

1. Establishment of stable inflammatory and immune cells expressing new IL-32α, β, γ or θ isoform.
 1) Promonocytic myeloma THP-1, KG-1a as well as lymphoma cell Jurkat expressing IL-32 isoforms.
 2) Search for genes such as cytokines and cell surface expressing genes modulated by IL-32

2. Preparation of IL-32mAbs for Confocal microscope, Immunohistochemistry, Western blot, and ELISA systems. IL-32 mAbs were also prepared for the relational study with related diseases (J. Immunol Method 333: 38; Proc Natl Acad Sci USA 103(9): 3298). In order to identify IL-32BP and regulated proteins, Proteomics/Yeast two hybrid system were also applied. IL-32 Tg mice were also established in order to study the function of a new cytokine (Oncogene 30: 3345, 2011; Cell Death Dis. 4: e640, 2013)

3. Interaction network mapping among IL-32 isoforms
 1) Comprehensive analyses to reveal 81 interactions between nine IL-32 isoforms (Biochimie 2014 Jan 25).
 2) IL-32δ interacted with IL-32β and inhibited IL-32β-mediated IL-10 production (FEBS Lett. 587: 3776, 2014)

4. Functional analysis of IL-32 isoforms mediated via their interacting factors in the isoform expressing cell lines.
 1) Intracellular association of IL-32α with PKCε and STAT3 augments IL-6 production in THP-1 promonocytic cells. J Biol Chem 287(42): 35556-64, 2012
 2) IL-32β-mediated C/EBPα phosphorylation by PKCδ abrogates the inhibitory effect of C/EBPα on IL-10 production. J Biol Chem 288 (33): 23650, 2013
